Michael O'Leary débute ses études au sein d'une institution catholique, le Clongowes Wood College. Il les poursuit au Trinity College de Dublin. Il en sortira sans diplôme.
De 1984 à 1986, il est conseiller fiscal chez KPMG. En 1988, il devient conseiller financier de Tony Ryan, le fondateur de Ryanair en 1985 avec Christy Ryan et Liam Lonergan. La compagnie aérienne régionale est alors fortement déficitaire. Au début, elle assure des vols entre Waterford, une ville du sud-est de l'Irlande, et Londres. Rapidement la compagnie élargit ses vols et propose des trajets intra européens.
Dans les années 1990, Tony Ryan se retire progressivement de la direction de Ryanair. Michael O'Leary devient PDG adjoint en 1991 puis PDG en janvier 1994. Il est chargé de restructurer l'entreprise afin d'augmenter sa rentabilité. Il mise sur un modèle d'avion unique, abandonne le concept de classe affaires et propose des tarifs très bas.
En 1992, il se rend à Dallas pour étudier le fonctionnement de Southwest Airlines. O'Leary transforme Ryanair, le petit transporteur, et en fait la première compagnie à bas coûts et la troisième compagnie aérienne d'Europe (après Air France/KLM et Lufthansa). Le siège social de Ryanair si situe à l'aéroport de Dublin.
La compagnie aérienne a connu une expansion rapide grâce à son modèle "low cost" et à la déréglementation de l'industrie du transport aérien en Europe en 1997.
En 2001, Michael O'Leary est nommé « homme d'affaires européen de l'année » par le magazine Fortune. Tony Ryan, le fondateur de Ryanair, décède en 2007 d'un cancer du pancréas.
