Ses parents sont instructeurs de plongée sous-marine au Club Méditerranée. Il passe son enfance près d'eux, entre la Grèce et l'ex-Yougoslavie. Il se tourne vers le cinéma suite à un accident qui l'empêche de poursuivre la plongée à haut niveau. Cette anecdote personnelle permet de mieux comprendre à quel point Le Grand Bleu, qu'il réalise en 1988, est une œuvre qui lui tient à cœur.

Il débute comme assistant réalisateur en France et réalise son premier long métrage en 1983. Intitulé Le Dernier Combat, il s'agit d'un film de science-fiction noir et blanc coécrit avec Pierre Jolivet. A 24 ans, sa distinction au Festival d'Avoriaz lui permet de signer un contrat avec la Gaumont. Sa carrière est lancée. Deux ans plus tard, il réalise Subway interprété notamment par Isabelle Adjani et Christophe Lambert. Le film est récompensé par trois oscars.

En 1988, Le Grand Bleu séduit le public, 10 millions de spectateurs se pressent dans les salles obscures. S'en suivront une série de blockbusters qui feront salle pleine : Nikita en 1990, Léon en 1994, Le cinquième élément en 1997, Jeanne d'Arc en 1999…

A la fin de la décennie 90, Luc Besson s'installe à Los Angeles avec son épouse, la comédienne Maïwenn Le Besco et leur fille Shanna. Il est décidé à conquérir le marché américain et Le cinquième élément marquera le début de cette carrière planétaire. Sa superproduction devient l'un des plus gros succès commerciaux d'un film français aux Etats-Unis. Pour couronner le tout, Luc Besson reçoit, en 1998, le César du meilleur réalisateur.

En 2000, il est président du jury au 53ème Festival de Cannes. C'est le début d'une nouvelle ère : Luc Besson crée sa société de production et de distribution, EuropaCorp et favorise la distraction et l'action. Le public est au rendez vous mais la critique dénonce le "système Besson" qui vise essentiellement la rentabilité financière en se basant toujours sur les mêmes scénarios : Taxi, Le transporteur, Yamakasi, ou encore Banlieue 13 en sont les principaux symboles…

Même si Luc Besson cherche à s'imposer sur le marché mondial, il met un point d'honneur à conserver le contrôle et la nationalité française de ses films d'un point de vue fiscal et juridique. C'est dans cette optique qu'il met en place le projet de création de la Cité du cinéma à Saint-Denis au début des années 2000.

Il revient à la réalisation en 2005 avec Angel-A, Arthur et les Minimoys et tout récemment Les aventures extraordinaires d'Adèle Blanc-Sec.

En avril 2007, l'autorité des marchés financiers donne son autorisation : la « Besson Incorporation » est cotée en bourse.

En 2008, Luc Besson crée l'agence de publicité « Blue », qui revendique être « la première agence française d'advertainment » (mélange entre la publicité et le divertissement). L'agence a notamment comme client l'UMP, parti pour lequel elle élabore une nouvelle plateforme de communication.

Il a réalisé certains clips musicaux d'Isabelle Adjani, de Serge Gainsbourg ou encore de Madonna. Il s'est également occupé de quelques films publicitaires, à l'instar de celui pour Dim en 1984, et de ceux pour l'Oréal.
